چکیده

We aimed to develop a reliable method based on Fourier transform infrared spectroscopy with attenuated total reflectance (FTIR-ATR) to discriminate Escherichia coli clones from B2(n = 9) and D(n = 13) phylogenetic groups. Eighty-eight E. coli isolates belonging to phylogenetic groups B2(n = 39) and D(n = 49), including particularly widespread high risk clones or clonal complexes (HiRCC) ST131, ST69, ST393 and ST405 were studied. Spectra were analysed by unsupervised (hierarchical cluster analysis-HCA) and supervised methods (soft independent modelling of class analogy-SIMCA and partial least square discriminant analysis-PLSDA). B2-ST131 isolates were discriminated from B2 non-ST131 and D phylogroup isolates (ST69, ST393, ST405) by HCA, SIMCA and PLSDA. D-ST69, D-ST393 and D-ST405 isolates were also distinguished from each other and from other STs from phylogroup D by the three methods. We demonstrate that FTIR-ATR coupled with chemometrics is a reliable and alternative method to accurately discriminate particular E. coli clones. Its validation towards an application at a routine basis could revolutionize high-throughput bacterial typing.

Graphene Oxide Antibacterial Sheets: Synthesis and Characterization (RESEARCH NOTE)

Graphene oxide (GO) was synthesized by oxidation of graphite powder using a time-saving modification of Hummers’ method and its antibacterial activity was investigated. Different techniques were applied to characterize the synthesized GO. X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM) and Atomic Force Microscopy (AFM) were used to investigate the crystallinity, morphology, topograp...

Cytotoxic and antimicrobial effects of biosynthesized ZnO nanoparticles using of Chelidonium majus extract

The basic goal of this study was to synthesize zinc oxide nanoparticles using the Chelidonium majus extract and asses their cytotoxic and antimicrobial properties. The synthesized ZnO NPs were characterized by UV-Vis, Scanning Electron Microscopy (SEM) with EDS profile, Fourier Transform Infrared Spectroscopy (FTIR), X-ray diffraction (XRD), Transmission Electron Microscopy (TEM) and Atomic For...

Phylogenic typing of Escherichia coli isolated from broilers with collibacillosis in Tabriz, North West of Iran

In this study, to know about the phylogeny of Escherichia coli isolated from broilers with collibacillosis in Tabriz, 70 E. coli isolates recovered from broilers with collibacillosis were characterized for phylogenetic group (A, B1, B2, D) by multiplex PCR. Of the all 70 samples, 35 (50%) isolates were classified as type A, 32 (45%) as type D, 2 (2.8%) as type B1 and 1 (2.8%) as type B2. This s...

Interaction of Cisplatin with Cellular Macromolecules: A Fourier Transform Infrared Spectroscopy Study

Platinum is a metallic element, which may react with our cellular component through its involvement in cancer chemotherapy medications. Cisplatin is one of the most useful antineoplastic drugs against human ovarian carcinoma, which has the central element of platinum in its structure. The nature of chemical interaction between platinum and cellular macromolecules is yet to be understood. We exa...

Rapid detection and identification of Pseudomonas aeruginosa and Escherichia coli as pure and mixed cultures in bottled drinking water using fourier transform infrared spectroscopy and multivariate analysis.

Fourier transform infrared (FT-IR) spectroscopy and multivariate analysis were used to identify Pseudomonas aeruginosa and Escherichia coli ATCC 25922 inoculated into bottled drinking water. Three inoculation treatments were examined: (i) E. coli ATCC 25922 (N = 3), (ii) P. aeruginosa (N = 3), and (iii) a 1:1 (v:v) mixed culture of both P. aeruginosa and E. coli ATCC 25922 (N = 3). The control ...
